چکیده

The objective of this present study was to isolate a potential laccase producing fungi from the different environmental samples from Gujarat. Eighteen morphologically different fungi were isolated on mineral salts medium having lignin (sole carbon source) and screened qualitatively for their ability to produce laccase by plate screening method using the indicators ABTS, Brilliant Blue R (RBBR), Syringaldazine and Guaicol. Six selected fungal isolates were further screened quantitatively in liquid medium and assayed using ABTS as a substrate. The highest laccase productivity in liquid medium was 166.2 U/ml (3 rd day) with BL-21/4. The genotypic characterization of selected fungal isolates was performed with partial 18S rRNA sequencing using ITS primer set ITS1/ITS4. The fungal isolates were identified as Aspergillus versicolor, Curvularia aeria, Curvularia lunata and Curvularia akaii. The conclusions of this study confirm that ABTS and Syringaldazine is very efficient phenolic compound for qualitative screening of novel ascomycetes that produces laccase.

Determination of lignin-modifying enzymes (LMEs) in Hyphodermella species using biochemical and molecular techniques

White-rot basidiomycetes are one of the most important lignolytic microorganisms. These fungi have been reported to secrete three main classes of lignin degrading enzymes: lignin peroxidases (LiPs), manganese peroxidases (MnPs) and laccases. In this study, for the first time the lignin degrading capability of two plant pathogens i.e. Hyphodermella rosae and H. corrugata was evaluated using both...

Laccase: Microbial Sources, Production, Purification, and Potential Biotechnological Applications

Laccase belongs to the blue multicopper oxidases and participates in cross-linking of monomers, degradation of polymers, and ring cleavage of aromatic compounds. It is widely distributed in higher plants and fungi. It is present in Ascomycetes, Deuteromycetes and Basidiomycetes and abundant in lignin-degrading white-rot fungi. It is also used in the synthesis of organic substance, where typical...

Occurrences, Physical and Biochemical Properties of Laccase

Laccases (benzenediol: oxygen oxidoreductase, EC 1.10.3.2) are multicopper oxidases that are widely distributed among plants, insects, and fungi. They belong to a group of polyphenol oxidases containing copper atoms in the catalytic centre which are usually called multicopper oxidases. It utilizes molecular oxygen to oxidize various aromatic and non-aromatic compounds. They have been described ...

Bioprospecting and biotechnological applications of fungal laccase.

Laccase belongs to a small group of enzymes called the blue multicopper oxidases, having the potential ability of oxidation. It belongs to enzymes, which have innate properties of reactive radical production, but its utilization in many fields has been ignored because of its unavailability in the commercial field. There are diverse sources of laccase producing organisms like bacteria, fungi and...

Exploring the Oxidation of Lignin-Derived Phenols by a Library of Laccase Mutants.

Saturation mutagenesis was performed over six residues delimiting the substrate binding pocket of a fungal laccase previously engineered in the lab. Mutant libraries were screened using sinapic acid as a model substrate, and those mutants presenting increased activity were selected for exploring the oxidation of lignin-derived phenols. The latter comprised a battery of phenolic compounds of int...
